چکیده

Situs inversus and polycystic kidney disease converge at primary cilia Although at first glance our body looks symmetrical, our internal organs obviously are arranged asymmetrically. It goes without saying that the heart is located on the left side and the liver on the right side yet such a seemingly trivial fact poses a very difficult biological question: How is the arrangement of the internal organs, the situs, regulated? Development of the organism starts with the fertilized oocyte, a round structure with no obvious asymmetry. At some point during development, however, axis formation occurs: A dorsal-ventral, an anterior-posterior and a leftright axis are established. Defects in left-right axis formation have been observed and are compatible with life, the patients suffer from situs inversus, i.e. their heart is located on the right side and the liver is located on the left side. As in many other circumstances, evidence obtained in the mouse has led to important insight into how the left-right axis may be for-

The Roles of Primary cilia in Polycystic Kidney Disease

Autosomal dominant polycystic kidney disease (ADPKD) is an inherited genetic disorder that results in progressive renal cyst formation with ultimate loss of renal function and other systemic disorders. These systemic disorders include abnormalities in cardiovascular, portal, pancreatic and gastrointestinal systems. ADPKD is considered to be among the ciliopathy diseases due to the association w...

The polycystic kidney disease proteins, polycystin-1, polycystin-2, polaris, and cystin, are co-localized in renal cilia.

Recent evidence has suggested an association between structural and/or functional defects in the primary apical cilium of vertebrate epithelia and polycystic kidney disease (PKD). In Caenorhabditis elegans, the protein orthologues of the PKD-related proteins, polycystin-1 (LOV-1), polycystin-2 (PKD2), and polaris (OSM-5), co-localize in the cilia of male-specific sensory neurons, and defects in...

Mouse models of polycystic kidney disease induced by defects of ciliary proteins

Polycystic kidney disease (PKD) is a common hereditary disorder which is characterized by fluid-filled cysts in the kidney. Mutation in either PKD1, encoding polycystin-1 (PC1), or PKD2, encoding polycystin-2 (PC2), are causative genes of PKD. Recent studies indicate that renal cilia, known as mechanosensors, detecting flow stimulation through renal tubules, have a critical function in maintain...

Cilia and polycystic kidney disease, kith and kin.

In the past decade, cilia have been found to play important roles in renal cystogenesis. Many genes, such as PKD1 and PKD2 which, when mutated, cause autosomal dominant polycystic kidney disease (ADPKD), have been found to localize to primary cilia. The cilium functions as a sensor to transmit extracellular signals into the cell. Abnormal cilia structure and function are associated with the dev...

New developments in the field of cystic kidney diseases.

For quite some time the field of polycystic kidney disease has led a life at the fringe of kidney research, but with the cloning of the PKD1 and many other genes this situation has dramatically changed. Polycystic kidney disease often is a syndromic disease affecting a variety of organs in addition to the kidney. Most of the proteins involved in polycystic kidney disease have been localized to ...
